Abstract: 5%~10% of the severe temporomandibular joint(TMJ) disorders are highly correlated to dysfunctional displacements and/or malformations of the TMJ disc, manifested as pathological disc attenuation, hyalinization, and perforation. Life quality of patient is affected by painful and dysfunctional joints. It is a great challenge to chose a safe, effective and replaceable TMJ disc implant for these patients. Tissue engineering is a promising approach for the creation of viable, effective TMJ disc functional implants. Recent studies on structural and biomechanical characterization of the TMJ disc and the principal components for the disc tissue- engineering approach, including scaffold, cell source, and biological/biomechanical stimuli were reviewed in this paper.
